I just met ezekiel who has a edu-pcr company 'amplyus'. They sell kits for
abt. $700 .
I don't know how much the parts required for openPCR cost , he said their
tech is similar.

I just did a phosphatase treatment of the cut vector. Now I want to

ligate

the two linear DNAs together.
Problem is, the phosphatase will digest the ATPs so that the ligase
won't ...more
P.S. the stop buffer is also not really a solution to the problem as it
will inactivate the ligase too. I would have to do an ethanol
precipitation
anyway...

Quick tip: if you are just buying time to think, don't freeze the DNA,
as freeze/thaw cycles damage DNA of all sizes (although the larger, the
worse the damage). So, just keep in the fridge unless ...more

usually you will put the product on a gel to confirm it's the DNA you
intended...?

or make sure the solution is at the optimal pH after you dilute with
water.
